Advancing In Vivo Solid Tumor Therapies

The strategic pivot focuses TScan’s efforts on developing in vivo-engineered TCR-T therapies, starting as singleplexed treatments and ultimately expanding into multiplexed options.

The company advanced two product candidates targeting PRAME and MAGE-A4 into IND-enabling studies.

By leveraging an in vivo engineering method, TScan aims to bypass lymphodepletion requirements while overcoming the high manufacturing costs and delivery delays typical of ex vivo therapies.

Management plans to release preclinical data in the first quarter of 2027, submit its initial IND application in the third quarter of 2027, and initiate Phase 1 development in the fourth quarter of 2027.

Phase 1 ALLOHA Trial Results and Strategic Pause

Capital constraints led TScan to pause enrollment in the Phase 3 ALLOHA-2 trial for TSC-101.

The business will continue tracking seven enrolled treatment-arm patients alongside 13 Cohort C patients from the Phase 1 ALLOHA trial at reduced costs.

In earlier Cohort A data, TSC-101 demonstrated durable remissions and lower relapse rates versus controls.

Recent Cohort C data revealed complete donor chimerism across all 13 tracked patients, including two who converted after receiving a third TSC-101 infusion or targeted agents.

Observed adverse events remained consistent with standard post-transplant profiles.

Preserving Value Through Strategic Partnerships

TScan expects to publish updated Cohort C data in the fourth quarter of 2026 and commercial-ready manufacturing process data in the second quarter of 2027.

The company intends to pursue strategic partnerships to preserve potential value across its heme and autoimmune programs.
